Protease enzymes, which break down proteins into peptides and amino acids, play a central role in digestion. The pancreas is the major source of digestive proteases in the human body, secreting enzymes such as trypsin, chymotrypsin, and elastase into the small intestine. Supplementation with protease is scientifically justified in cases where pancreatic function is compromised, such as in exocrine pancreatic insufficiency (EPI), chronic pancreatitis, or cystic fibrosis. In these conditions, the pancreas fails to secrete adequate digestive enzymes, leading to malabsorption and nutritional deficiencies. Clinical studies show that pancreatic enzyme replacement therapy (PERT), which includes proteases, improves nutrient absorption, reduces gastrointestinal symptoms, and enhances quality of life in affected patients (Lowe et al., 2014).
For individuals with normal pancreatic function, there is limited evidence that supplemental proteases further "support" the pancreas or enhance its health. Most scientific validation supports protease supplementation in the context of enzyme deficiency rather than general pancreatic health maintenance. Nevertheless, the link between protease supplementation and pancreatic support is well established in the context of enzyme insufficiency, earning it a relatively high evidence rating.
