Zinc has been investigated as a potential adjunctive treatment in schizophrenia based on emerging scientific evidence rather than traditional use. Several studies have noted that patients with schizophrenia often exhibit lower serum zinc levels compared to healthy controls. Zinc plays a key role in modulating neurotransmitter systems, including glutamatergic and GABAergic transmission, both of which are implicated in the pathophysiology of schizophrenia. Some small-scale clinical trials have examined zinc supplementation (usually in addition to antipsychotic medication) and reported modest improvements in negative symptoms and overall clinical outcomes. For example, a randomized, double-blind, placebo-controlled trial published in Biological Psychiatry (2009) found that zinc supplementation as an adjunct to atypical antipsychotics improved negative symptoms more than placebo. However, these studies are limited by small sample sizes and short durations. Meta-analyses and systematic reviews suggest that while there is some promise, the quality of evidence is low to moderate, and more research is needed to establish the efficacy and optimal dosing of zinc for schizophrenia. Overall, the use of zinc is supported by preliminary scientific data but is not yet a standard or widely endorsed treatment, and the evidence remains insufficient for strong recommendations.
