Zinc is an essential trace element that plays a significant role in bone metabolism, making it of interest in the context of osteoporosis. Scientific studies have established that zinc is a cofactor for numerous enzymes involved in bone formation and mineralization. Experimental research in animals and in vitro studies have shown that zinc stimulates osteoblastic bone formation and inhibits osteoclastic bone resorption, which are beneficial effects for bone health. Epidemiological studies have reported associations between low dietary zinc intake or low serum zinc levels and reduced bone mineral density (BMD) in various populations, particularly postmenopausal women who are at increased risk of osteoporosis.
However, while these findings suggest a potential role for zinc in maintaining bone health, the direct evidence from randomized controlled trials (RCTs) in humans using zinc supplementation to prevent or treat osteoporosis is limited and inconclusive. Most clinical studies are small, have methodological limitations, or combine zinc with other nutrients like calcium and vitamin D, making it difficult to isolate the effect of zinc alone. Systematic reviews generally conclude that while zinc is necessary for bone health, there is insufficient high-quality evidence to support zinc supplementation as an effective standalone treatment or preventive measure for osteoporosis. Thus, zinc’s use for osteoporosis is scientifically plausible and supported by mechanistic and epidemiological data, but clinical evidence for supplementation is weak.
