Selenium is a trace mineral with a well-established role in human health, primarily through its incorporation into selenoproteins, which are crucial for antioxidant defense and redox regulation. Within the context of mitochondrial health, selenium supports the function of several enzymes, most notably glutathione peroxidases and thioredoxin reductases. These selenoproteins help to neutralize reactive oxygen species (ROS) generated during mitochondrial energy production, thereby protecting mitochondrial membranes and DNA from oxidative damage. Scientific studies have shown that selenium deficiency leads to impaired mitochondrial function, increased oxidative stress, and cellular damage. Animal models and in vitro studies have demonstrated that adequate selenium levels can help maintain mitochondrial integrity and function. However, direct clinical trials specifically targeting selenium supplementation for mitochondrial diseases in humans are limited; most evidence comes from mechanistic studies, observational data, or research in other oxidative stress-related conditions. Therefore, while the biochemical rationale for selenium's support of mitochondrial health is strong and supported by preclinical evidence, robust clinical validation is still developing. Selenium remains an essential nutrient for mitochondrial antioxidant defense, but its targeted use in mitochondrial disorders should be evidence-based and cautious, given both the potential benefits and risks of supplementation.
