Zinc is a trace mineral with multiple physiological roles, including involvement in antioxidant defense, neurotransmitter regulation, and modulation of inflammation—all processes that may be relevant to migraine pathophysiology. Recent scientific interest has focused on the potential association between zinc status and migraine. Several observational studies have reported lower serum zinc levels in migraine sufferers compared to healthy controls. Randomized controlled trials (RCTs) are limited but a few small studies have suggested that zinc supplementation, often at doses of 15-30 mg per day, may lead to a reduction in headache frequency, severity, or associated symptoms in some patients. The proposed mechanisms include zinc’s anti-inflammatory and antioxidant effects, as well as its influence on neuronal excitability and neurotransmitter balance (notably glutamate and gamma-aminobutyric acid regulation). However, the overall quality of evidence remains low. Most studies are small, of short duration, and sometimes combine zinc with other nutrients, making it difficult to isolate its effects. Systematic reviews and meta-analyses highlight promising but preliminary results, and leading headache societies do not currently recommend zinc supplementation as a standard treatment for migraine. In conclusion, while there is emerging scientific interest and some supportive data, the evidence is not yet robust enough to firmly endorse zinc for migraine management outside of research settings or confirmed deficiency.
