Punicalagins are ellagitannin compounds predominantly found in pomegranate (Punica granatum) and have been studied for their potential benefits in oral health, particularly for conditions like gingivitis. Scientific interest in punicalagins for gingivitis stems from their strong antioxidant and antimicrobial properties. In vitro studies have demonstrated that punicalagins can inhibit the growth of oral pathogens associated with gingivitis, such as Porphyromonas gingivalis and Streptococcus mutans. Additionally, punicalagins have been shown to reduce inflammation and oxidative stress in gingival tissues.
Several small clinical trials and randomized controlled studies using pomegranate extracts (which are rich in punicalagins) as mouthwash or gel have reported reductions in plaque accumulation, gingival bleeding, and markers of inflammation compared to placebo or chlorhexidine. However, these studies often use whole pomegranate extracts, making it difficult to attribute effects solely to punicalagins. The quality and size of available human studies are moderate, and more robust clinical trials are needed to firmly establish efficacy.
