Ananain is a cysteine protease enzyme derived from the stem of the pineapple plant (Ananas comosus). Its use in treating or supporting gingivitis is based on its proteolytic (protein-digesting) activity, which can help degrade necrotic tissue and reduce biofilm formation in the oral cavity. Several in vitro studies have shown that enzymes like ananain and bromelain can disrupt dental plaque and may have mild anti-inflammatory properties. A handful of small-scale clinical studies and pilot trials have indicated a potential benefit for reducing gingival inflammation and plaque index scores when enzyme-containing mouthwashes or gels are used adjunctively with regular oral hygiene.
However, the clinical evidence remains limited and of low quality. Most studies have been short-term, involved small sample sizes, and often combined ananain with other enzymes such as bromelain or papain, making it difficult to isolate its specific effect. Systematic reviews have not established a strong consensus for the routine use of such enzymes in managing gingivitis, and major dental organizations do not currently recommend ananain as a primary therapy. In summary, while there is some emerging scientific evidence for its adjunctive benefit, it is not robust, and more well-designed clinical trials are needed.
