Polygala root (Polygala tenuifolia), known as Yuan Zhi in traditional Chinese medicine (TCM), has been historically used to calm the mind, nourish the heart, and treat symptoms related to mood disturbances such as anxiety, insomnia, and what might be interpreted as depression. Ancient TCM texts describe its use for “shen” (spirit) disorders, which overlap conceptually with modern understandings of depression. Contemporary scientific research is limited but growing; a few animal studies suggest that extracts of polygala root may have antidepressant-like effects, possibly by modulating neurotransmitters such as serotonin or dopamine, or by reducing inflammation and oxidative stress in the brain. However, robust clinical trials in humans are lacking, and the evidence is not yet strong enough to recommend polygala root as a mainstream treatment for depression. The rating of 2 reflects a modest body of preclinical work and considerable traditional use, but insufficient high-quality clinical data. As with many herbal remedies, more research is needed to determine its efficacy and safety for depression in humans.
