Chalcone is a naturally occurring compound found in many plants and is a precursor for various biologically active flavonoids. There is emerging scientific interest in chalcone and its derivatives due to their antimicrobial properties. Several in vitro studies have demonstrated that chalcone compounds exhibit antibacterial activity against both Gram-positive and Gram-negative bacteria, including drug-resistant strains. Proposed mechanisms include disruption of bacterial cell walls, inhibition of efflux pumps, and interference with nucleic acid synthesis. However, most of the evidence comes from cell culture studies or, less frequently, animal models; there are currently no robust human clinical trials validating chalcone as a direct alternative to conventional antibiotics.
Chalcone is not traditionally used as an "antibiotic alternative" in herbal or folk medicine, but its antimicrobial potential has attracted attention in medicinal chemistry as a lead compound for novel antibiotic development. The rating is 2 because, while there is promising preclinical evidence and a plausible mechanism of action, there is insufficient clinical evidence to support its use as a replacement for standard antibiotics in humans.
