Osthole is a natural coumarin compound predominantly found in the fruit of Cnidium monnieri, a traditional Chinese medicinal herb. Its use for supporting the skeletal system, particularly bone health, has some scientific basis, largely from preclinical research. Studies in cell cultures and animal models have demonstrated that osthole can stimulate osteoblast proliferation and differentiation, which are critical processes for bone formation. It has also been shown to inhibit osteoclast activity, the cells responsible for bone resorption. These dual actions suggest a potential to prevent or treat osteoporosis. One frequently cited mechanism involves the upregulation of bone morphogenetic proteins (BMPs) and Wnt/β-catenin signaling pathways, both essential for bone growth and remodeling. In ovariectomized rat models (a common model for postmenopausal osteoporosis), osthole treatment improved bone mineral density and microarchitecture.
However, it is important to note that there is a lack of high-quality human clinical trials confirming these effects. Most of the available evidence comes from in vitro and animal studies. While osthole has a long history of use in traditional Chinese medicine for bone-related disorders, its scientific validation is still emerging. In summary, osthole shows promise as a bone-supporting agent based on preclinical studies, but more rigorous human research is needed to firmly establish its efficacy and safety for skeletal health.
