The historical basis for its use comes primarily from clinical practice in the mid-20th century, when chlorophyllin was included in ointments and mouth rinses for various wounds and ulcers. Several small studies and case reports from the 1940s to 1960s suggested that chlorophyllin might aid healing and reduce pain in oral ulcers, but these were not large, well-controlled studies by modern standards.

The proposed mechanisms for chlorophyllin’s effect include its purported anti-inflammatory, antimicrobial, and deodorizing properties, which might contribute to a more favorable environment for mucosal healing. However, there is little contemporary scientific research specifically addressing chlorophyllin’s effectiveness for canker sores. Systematic reviews and modern clinical guidelines do not mention chlorophyllin as a recommended treatment, and there is a lack of robust, high-quality clinical trials validating its use for this specific indication.
