Synopsis

Source of validity: Scientific
Rating (out of 5): 2

Urolithin A is a metabolite derived from ellagitannins found in foods like pomegranates, berries, and nuts. Its relevance to skin health, particularly in the context of wrinkles, stems from its demonstrated effects on mitochondrial function, cellular health, and anti-inflammatory pathways. Preclinical studies (in vitro and animal models) have shown that Urolithin A can promote mitophagy (the selective recycling of damaged mitochondria), reduce oxidative stress, and modulate inflammation—all factors implicated in skin aging and the formation of wrinkles. For example, research published in Nature Metabolism and other journals has demonstrated Urolithin A’s ability to enhance mitochondrial health and cellular function, which may theoretically translate to healthier, more youthful skin. Additionally, Urolithin A has been shown to upregulate collagen synthesis and inhibit matrix metalloproteinases (MMPs), enzymes that break down collagen and contribute to wrinkle formation, in cell-based studies.

However, direct clinical studies in humans specifically investigating Urolithin A’s efficacy for reducing wrinkles are very limited. Most human trials to date have focused on systemic effects such as muscle health and mitochondrial function rather than visible skin aging. As such, while the biological plausibility and early mechanistic evidence are promising, robust clinical validation in the context of wrinkles is still lacking. The current evidence level is rated as 2/5, reflecting preclinical promise but insufficient direct human data.
