The primary reason for its use is based on strong scientific evidence: isoniazid can cause peripheral neuropathy as a side effect, and vitamin B6 supplementation significantly reduces this risk. Isoniazid interferes with vitamin B6 metabolism, leading to a functional deficiency that manifests as nerve damage. Numerous clinical studies and treatment guidelines from organizations such as the World Health Organization (WHO) and the Centers for Disease Control and Prevention (CDC) recommend vitamin B6 supplementation (typically 10-50 mg daily) for patients on isoniazid, especially those at higher risk of neuropathy (e.g., malnourished, pregnant women, those with HIV, alcoholics, or diabetics).
