Androstenedione is a steroid hormone that serves as a direct precursor to both testosterone and estrogen in the human body. Its use as a supplement to treat or support low testosterone is based on the scientific rationale that increasing this precursor might increase testosterone levels. Early laboratory studies and some small clinical trials have shown that androstenedione supplementation can transiently raise serum testosterone levels in some men. However, multiple larger, well-controlled studies have failed to show consistent or clinically meaningful improvements in testosterone concentrations, muscle strength, or other markers of androgenic activity in healthy young or older men. Furthermore, androstenedione supplementation has been associated with increased estrogen levels, which may be undesirable in men, and potential adverse effects such as altered lipid profiles and changes in liver enzymes. The use of androstenedione is banned by most sports organizations due to its potential for performance enhancement and health risks.
