7,8-Dihydroxyflavone (7,8-DHF) is a synthetic flavonoid that has attracted scientific interest due to its action as a potent and selective agonist of the TrkB receptor, the primary receptor for brain-derived neurotrophic factor (BDNF). Preclinical studies, mostly in animal models, have demonstrated that 7,8-DHF can mimic BDNF's neuroprotective and neurotrophic effects, leading to improved neuronal survival and synaptic plasticity. Since chronic stress is associated with reduced BDNF signaling and atrophy in brain regions such as the hippocampus, 7,8-DHF has been investigated for its potential to counteract these stress-related changes.
In rodent studies, administration of 7,8-DHF has been shown to reduce behavioral and physiological markers of stress. For example, mice treated with 7,8-DHF exhibited reduced anxiety-like and depressive-like behaviors in standard behavioral paradigms after exposure to chronic stress. The compound also prevented stress-induced reductions in BDNF signaling and synaptic markers in the brain. However, it is important to note that as of 2024, evidence in humans is lacking—there are no published clinical trials assessing the efficacy or safety of 7,8-DHF for stress or stress-related disorders.
