Siphoviridae is a family of bacteriophages—viruses that infect bacteria. Their use against Staphylococcus infections, particularly those caused by Staphylococcus aureus (including MRSA), is grounded in scientific research. Bacteriophage therapy has been studied since the early 20th century, but interest has resurged in recent decades due to rising antibiotic resistance. Multiple in vitro and animal studies have demonstrated that certain Siphoviridae phages can specifically infect and lyse Staphylococcus species. For example, phages from the Siphoviridae family have been shown to reduce bacterial loads and improve outcomes in animal models of staphylococcal infections, including wound and systemic infections.

Clinical trials in humans are more limited but growing; some early-phase studies and case reports suggest promising safety and efficacy profiles for phage preparations containing Siphoviridae members, often as part of personalized phage cocktails. These findings are supported by genomic analyses that confirm these phages' specificity for Staphylococcus and lack of deleterious genes. The evidence base is not yet as robust as for conventional antibiotics, but ongoing research is strengthening the scientific rationale for their use, especially for antibiotic-resistant strains. Thus, while more high-quality clinical trials are needed, the current use of Siphoviridae-based phage therapy for staph infections is grounded in scientific evidence.
