5-Methoxytryptamine (5-MT), also known as mexamine, is an endogenous indoleamine structurally related to both serotonin and melatonin. Its use for supporting or treating sleep disturbances is justified by scientific research, although evidence is moderate and primarily derived from preclinical studies. 5-MT is a naturally occurring compound in the pineal gland and is a metabolic intermediate in the biosynthesis of melatonin, a well-established regulator of circadian rhythms and sleep. Some animal studies demonstrate that administration of 5-MT can produce sedative and sleep-promoting effects, likely by acting on melatonin and serotonin receptors (MT1, MT2, 5-HT receptors). For example, rodent models have shown increased non-REM sleep and shortened sleep latency with 5-MT administration. Additionally, 5-MT has been shown to exhibit chronobiotic properties, influencing sleep-wake cycles. However, there is limited clinical evidence in humans; most data are from in vitro or animal research, and direct studies on sleep-disturbed individuals are lacking. While the mechanistic rationale is strong—given its place in melatonin biosynthesis and receptor activity—clinical trials specifically evaluating its efficacy for restless or disturbed sleep in people are needed. In summary, while there is a scientific basis supporting the potential role of 5-MT in sleep regulation, the quality and quantity of evidence remain moderate, with more research required to establish efficacy and safety in humans.
