Transforming growth factor beta (TGF-β) has scientific validation for its role in supporting the skeletal system, particularly bone development and remodeling. TGF-β is a multifunctional cytokine that regulates cellular proliferation, differentiation, and apoptosis. In the context of bone health, numerous studies have demonstrated that TGF-β is critical in the regulation of osteoblast (bone-forming cell) and osteoclast (bone-resorbing cell) activity. It stimulates the proliferation and differentiation of osteoprogenitor cells, enhances the production of extracellular matrix proteins, and plays a key role in the coupling of bone formation and resorption during bone remodeling.

Experimental models have shown that mice lacking TGF-β signaling in bone cells exhibit impaired bone formation, reduced bone mass, and abnormal bone architecture. Moreover, TGF-β is involved in fracture healing by recruiting mesenchymal stem cells and promoting their differentiation into chondrocytes and osteoblasts. Human studies, while more limited, have also suggested that dysregulation of TGF-β pathways can contribute to bone diseases such as osteoporosis and osteogenesis imperfecta.

However, it should be noted that TGF-β is not typically used as a direct supplement in humans due to its complex and context-dependent effects. Rather, its role in bone health is well-established at the molecular and cellular levels through scientific research, supporting its importance in the maintenance and repair of the skeletal system.
