Platelet-derived growth factor (PDGF) is scientifically validated as a key regulator in bone repair and regeneration. PDGF is a potent mitogen for cells of mesenchymal origin, including osteoblasts and fibroblasts, which are critical in bone healing. Numerous preclinical studies and clinical trials have demonstrated that PDGF, particularly in recombinant form (e.g., becaplermin), enhances bone regeneration by stimulating cellular proliferation, chemotaxis, angiogenesis, and extracellular matrix formation.
In orthopedic and dental applications, PDGF has been incorporated into bone graft materials and used in procedures such as spinal fusion and periodontal regeneration. For example, recombinant human PDGF-BB (rhPDGF-BB) has been approved by the FDA for use in conjunction with bone grafts to treat chronic foot ulcers and is also used in bone defect repair and oral/maxillofacial surgery. Clinical studies have shown improved outcomes in bone defect closure, new bone formation, and periodontal tissue regeneration when PDGF is applied locally at the site of injury or surgery.
However, while the evidence is robust for local application in surgical or wound-healing contexts, there is limited evidence supporting systemic use of PDGF for general bone health. Thus, the use of PDGF in supporting the skeletal system is well-justified in specific therapeutic settings, particularly for enhancing localized bone regeneration and repair.
