MK-677, also known as Ibutamoren, is a non-peptide agonist of the ghrelin receptor and is known for its ability to increase growth hormone (GH) and insulin-like growth factor 1 (IGF-1) levels. Scientific studies have investigated its effects on bone metabolism, as GH and IGF-1 are both critical for bone growth and remodeling. Clinical research, including randomized, double-blind, placebo-controlled trials in elderly adults, has shown that MK-677 administration increases markers of bone formation and bone mineral density (BMD) over periods of up to 12 months. For instance, one study published in the journal "The Journal of Clinical Endocrinology & Metabolism" (2001) demonstrated that MK-677 reverses age-related growth hormone decline and increases BMD in elderly subjects, suggesting a potential therapeutic role for osteoporosis or other bone-wasting conditions.

However, while these results are promising, long-term data on fracture risk reduction or definitive prevention of osteoporosis with MK-677 are lacking. The majority of evidence comes from short- to medium-term studies with relatively small sample sizes. There are also potential side effects, such as increased appetite and insulin resistance, that warrant caution. Thus, while there is scientific validation for the use of MK-677 to support the skeletal system, particularly through its effects on bone turnover markers and BMD, the overall strength of evidence is moderate, not definitive.
