Ibutamoren (MK-677) is an orally active, non-peptide agonist of the ghrelin receptor that stimulates the secretion of growth hormone (GH) and insulin-like growth factor 1 (IGF-1). There is scientific evidence supporting its potential benefit to the skeletal system, particularly in the context of bone health. Several clinical studies have shown that Ibutamoren increases circulating GH and IGF-1 levels, hormones known to play a critical role in bone formation and remodeling. For example, a randomized, placebo-controlled trial in elderly adults found that daily administration of Ibutamoren increased bone turnover markers and IGF-1 concentrations, suggesting enhanced bone remodeling activity (Murphy et al., 1998). Another study in healthy obese males also demonstrated increased bone formation markers with Ibutamoren treatment (Nass et al., 1999).

While these results are promising and mechanistically plausible, there is limited long-term data demonstrating actual increases in bone mineral density or reductions in fracture risk. Most research to date has focused on surrogate markers such as biochemical markers of bone turnover rather than direct clinical outcomes. Therefore, while there is scientific rationale and early evidence suggesting Ibutamoren may support bone health, more robust clinical trials are needed to confirm its effectiveness in improving skeletal outcomes.
