Hydroxyproline is an amino acid found almost exclusively in collagen, a primary structural protein in bone, skin, and connective tissue. Its presence is crucial for the stability of the collagen triple helix structure. Scientific evidence supports the association between hydroxyproline and bone health primarily because hydroxyproline serves as a biomarker for collagen turnover and bone metabolism. Studies have shown that oral collagen supplementation, which contains hydroxyproline, may contribute to improved bone mineral density and support bone repair, particularly in postmenopausal women and individuals with osteoporosis. The rationale is that dietary collagen peptides, rich in hydroxyproline, may stimulate osteoblast activity and collagen synthesis in bone tissue, thus supporting skeletal integrity. However, while there is evidence that hydroxyproline intake via collagen supplements influences collagen metabolism, direct evidence linking isolated hydroxyproline supplementation (as opposed to whole collagen or gelatin) to improved bone health in humans is limited. Most clinical studies focus on collagen peptides rather than hydroxyproline alone. Therefore, while the role of hydroxyproline as a marker and component of collagen is scientifically validated, the strength of evidence for its direct use as a supplement to support the skeletal system rates as moderate.
