Pyridoxal-5-phosphate (P-5-P) is the active coenzyme form of vitamin B6 and plays a crucial role in the synthesis of neurotransmitters, including serotonin. P-5-P serves as a cofactor for the enzyme aromatic L-amino acid decarboxylase, which converts 5-hydroxytryptophan (5-HTP) to serotonin. Without sufficient P-5-P, this enzymatic reaction is impaired, potentially leading to suboptimal serotonin production.
There is scientific evidence from biochemical and clinical studies supporting the role of vitamin B6 (and specifically P-5-P) in neurotransmitter synthesis. Several studies have demonstrated that vitamin B6 deficiency can lead to decreased levels of serotonin and other neurotransmitters, contributing to mood disorders such as depression. Supplementation with P-5-P or vitamin B6 has been shown to restore normal neurotransmitter levels in deficient individuals.
However, in individuals with adequate vitamin B6 status, additional supplementation with P-5-P may not significantly boost serotonin levels. The evidence supporting its use for mood or serotonin support in the general population is moderate, with stronger support in cases of deficiency or genetic conditions affecting B6 metabolism. Thus, while there is a clear biochemical rationale and some clinical evidence, the overall strength of evidence for routine serotonin support is moderate.
