Nerve Growth Factor (NGF) is a neurotrophin important for the growth, maintenance, and survival of certain neurons. Its role in neurodegenerative diseases, including conditions associated with "senility" such as Alzheimer's disease, has been studied since the late 20th century. Preclinical research in animal models has shown that NGF administration can promote neuronal survival and improve cognitive performance, suggesting a potential therapeutic role for age-related cognitive decline. In humans, small clinical trials and early-phase studies have tested NGF delivery—often via intranasal administration or gene therapy—in patients with Alzheimer's disease or mild cognitive impairment. These studies have demonstrated some biochemical and neuroimaging changes consistent with NGF activity, and a few have suggested modest cognitive benefits or delayed progression of symptoms. However, these trials are limited by small sample sizes, short durations, and challenges related to NGF delivery (such as blood-brain barrier penetration and side effects like pain).
