7,8-Dihydroxyflavone (7,8-DHF) is a small molecule that acts as a selective agonist of the TrkB receptor, which is the main receptor for brain-derived neurotrophic factor (BDNF). The rationale for its use in Post Traumatic Stress Disorder (PTSD) is based on preclinical research showing that dysregulation of BDNF/TrkB signaling may be involved in the pathophysiology of PTSD. Animal studies, particularly in rodents, have shown that administration of 7,8-DHF can enhance extinction of conditioned fear responses and reduce anxiety-like behaviors, which are relevant to PTSD symptoms. For example, studies published in journals such as Translational Psychiatry and Journal of Neuroscience (Andero et al., 2011; Johnson et al., 2016) have demonstrated that 7,8-DHF facilitates fear extinction and may reverse stress-induced synaptic deficits in the amygdala and prefrontal cortex—key brain regions implicated in PTSD. However, there have been no published clinical trials in humans as of mid-2024, and evidence is limited to animal models and mechanistic studies. Thus, while the scientific rationale is plausible and the preclinical evidence is promising, the therapeutic use of 7,8-DHF for PTSD in humans remains unproven, and its safety and efficacy have not been established in clinical settings.
