Vitamin B12 (specifically in its active forms such as 5-deoxyadenosylcobalamin and methylcobalamin) is scientifically validated as both a cause and a treatment for pernicious anemia. Pernicious anemia is an autoimmune disorder characterized by the inability to absorb vitamin B12 due to lack of intrinsic factor, a protein necessary for B12 absorption in the gut. The disease leads to megaloblastic anemia and neurological symptoms if untreated. The connection between B12 deficiency and pernicious anemia was established in the early 20th century. Early treatments involved consuming large amounts of liver, which is rich in B12, and later, direct B12 supplementation became the standard of care once the vitamin was isolated and synthesized.

Numerous clinical studies and decades of medical practice have confirmed that B12 supplementation (in injectable or high oral doses) corrects the hematological and neurological manifestations of pernicious anemia. The use of 5-deoxyadenosylcobalamin is justified by the fact that it is one of the biologically active forms of B12 in human metabolism. Current medical guidelines universally recommend B12 supplementation in cases of pernicious anemia, and failure to treat leads to irreversible neurological damage and death. Thus, the use of vitamin B12 for pernicious anemia is one of the most well-established and scientifically validated treatments in medicine.
