Beta-caryophyllene is a natural bicyclic sesquiterpene found in many essential oils, especially clove oil, black pepper, and cannabis. Scientific research supports its use for general pain relief, primarily due to its unique pharmacological action as a selective agonist of the cannabinoid receptor type 2 (CB2). Unlike CB1 receptors (which are involved in the psychoactive effects of cannabinoids), CB2 receptors are mainly expressed in peripheral tissues and immune cells and are involved in modulating inflammation and pain.

Preclinical studies in animal models have demonstrated that beta-caryophyllene can reduce pain responses in various types of pain, including inflammatory and neuropathic pain. For example, research published in European Journal of Pharmacology (2007) showed that beta-caryophyllene significantly reduced inflammatory pain in mice via CB2 receptor activation. Other studies have reported similar analgesic effects in models of neuropathic pain, with the mechanism mainly attributed to its anti-inflammatory properties and modulation of the endocannabinoid system.

However, while the preclinical evidence is robust, clinical studies in humans are still limited. Most of the scientific support comes from animal models, with only a few small-scale human studies or case reports suggesting analgesic effects. Therefore, while the scientific rationale is strong and the mechanism is well-understood, further clinical research is needed to confirm its efficacy and safety in humans for general pain management.
