Several observational studies have found an association between low vitamin D levels and symptoms such as fatigue, low mood, and cognitive difficulties. There is some biological plausibility for vitamin D’s involvement, as vitamin D receptors are found in brain tissue, and the vitamin is thought to influence neurotrophic factors, neurotransmitter synthesis, and inflammatory pathways that may impact nervous system function.

However, direct evidence for vitamin D as a treatment for “nervous exhaustion” is limited. Most intervention studies focus on vitamin D's effects on depression, fatigue, or general well-being rather than the historical syndrome of nervous exhaustion. Meta-analyses suggest a modest benefit of vitamin D supplementation in individuals with clinically significant deficiency, particularly for depressive symptoms, but results are inconsistent and effect sizes are generally small.
