Phosphatidylethanolamine (PE) is a major phospholipid component of biological membranes, including those in the nervous system. Scientific interest in PE’s role in neural health stems from its abundance in neuronal membranes and its involvement in membrane fluidity, vesicle formation, and cell signaling. Research indicates that PE contributes to the structural integrity and function of synaptic membranes and is crucial for proper neurotransmitter release. Deficiencies or disruptions in PE metabolism have been linked to neurodegenerative conditions in animal models, such as Parkinson’s disease and Alzheimer’s disease, where altered phospholipid composition correlates with disease pathology. However, direct clinical evidence supporting the supplementation of PE for nerve health in humans remains limited. Most studies focus on the molecular and cellular level, highlighting the essential nature of PE in maintaining nervous system structure and function. Some preclinical studies suggest that increasing PE levels can ameliorate neural dysfunction in models of neurodegeneration. Nevertheless, robust clinical trials demonstrating clear benefits of oral or supplemental PE in supporting nerve function in humans are lacking. In conclusion, while there is a solid mechanistic basis and some preclinical evidence, the direct clinical validation for PE supplementation in supporting the nervous system is still emerging.
