Nicotinamide riboside (NR) is a form of vitamin B3 and a precursor to nicotinamide adenine dinucleotide (NAD+), a molecule involved in cellular energy metabolism and repair. Interest in NR for nerve damage arises from preclinical research indicating that boosting NAD+ levels can support nerve cell health, promote axonal regeneration, and potentially reduce neurodegeneration. Animal studies have shown that NR supplementation can increase NAD+ levels in nervous tissue and may reduce nerve damage in models of chemotherapy-induced peripheral neuropathy, diabetic neuropathy, and traumatic nerve injury. Additionally, NR has demonstrated the ability to enhance mitochondrial function and reduce oxidative stress in nerve cells.

However, human evidence is currently limited. Few small clinical trials have explored NR’s effects on peripheral neuropathy or neurodegeneration, and most have only assessed NAD+ bioavailability or safety rather than direct nerve repair. Larger, high-quality randomized controlled trials are needed to confirm clinical benefits in humans.
