Ibutamoren (also known as MK-677) is a non-peptide agonist of the ghrelin receptor and stimulates the secretion of growth hormone (GH) and insulin-like growth factor 1 (IGF-1). There is scientific evidence, primarily from small clinical trials and animal studies, to suggest that Ibutamoren can improve muscle mass and muscle strength in some populations. For example, a double-blind, placebo-controlled trial in healthy elderly adults showed that daily administration of Ibutamoren increased lean body mass after 2 months of treatment (Annals of Internal Medicine, 2008). Another study in older adults found improvements in GH and IGF-1 levels, which are hormones associated with muscle protein synthesis and muscle maintenance.

However, while these findings are promising for lean mass, the evidence directly linking Ibutamoren to improvements in "muscle tone" (a term referring more specifically to the resting tension and firmness of muscles, not just mass) is limited. Most clinical studies have focused on muscle mass or function rather than tone per se. The safety profile of Ibutamoren is still under investigation, and it is not approved for this use by regulatory agencies. Thus, while there is scientific rationale and some supporting evidence for its ability to support muscle mass (which can indirectly influence muscle tone), the quality and quantity of evidence are moderate, justifying a rating of 3 out of 5.
