Guanidinopropionic Acid (GPA) is a creatine analog that has been investigated primarily in preclinical studies for its effects on cellular energy metabolism, particularly in relation to mitochondrial function. GPA competes with creatine for uptake and utilization, leading to a reduction in phosphocreatine stores and a compensatory upregulation of mitochondrial biogenesis and oxidative capacity in muscle cells. This has been demonstrated in rodent models, where GPA administration increased the expression of proteins involved in mitochondrial energy metabolism and promoted a shift from glycolytic to oxidative muscle fibers. Some studies have also reported enhanced glucose uptake and improved insulin sensitivity after GPA treatment, potentially linked to its mitochondrial effects.

However, it is important to note that most evidence supporting GPA’s impact on mitochondria comes from animal or in vitro models. There is a lack of robust clinical trials in humans assessing its safety and efficacy for supporting mitochondrial health. Furthermore, GPA is not widely used as a dietary supplement or therapeutic agent for mitochondrial support in humans, and its use is not established in traditional medicine systems. Therefore, while there is scientific rationale and moderate preclinical evidence for GPA’s role in modulating mitochondrial function, the evidence base is currently limited and does not support routine use in humans for this purpose.
