Alpha-ketoglutarate (AKG) plays a central role in cellular metabolism as a key intermediate of the Krebs (citric acid) cycle, which is crucial for mitochondrial energy production. This biochemical rationale underpins its use for mitochondrial support. Scientific studies in cell cultures and animal models have shown that AKG supplementation can enhance mitochondrial function, support ATP production, and reduce markers of oxidative stress. For instance, research in rodents reports that AKG can extend lifespan and improve age-related declines in mitochondrial function (Cell Metabolism, 2020). Additionally, AKG has been shown to act as an antioxidant and modulate cellular signaling pathways relevant to mitochondrial health. However, robust clinical evidence in humans is limited. A few small-scale human studies suggest potential benefits in metabolic health and muscle function, but these are not specifically focused on mitochondrial outcomes and are often confounded by other interventions.
