Niacinamide (vitamin B3) has some scientific rationale for its use in supporting memory and brain function, but the evidence is limited and primarily indirect. Niacinamide is a precursor to NAD+ (nicotinamide adenine dinucleotide), a vital coenzyme involved in cellular energy production and DNA repair. Adequate levels of NAD+ are essential for neuronal health and function. Severe deficiency of vitamin B3 (as seen in pellagra) can cause neurological symptoms such as confusion, memory loss, and dementia, which are reversed by supplementation. This forms the foundation for considering niacinamide important for brain health.
However, in individuals with normal niacin status, there is little robust clinical evidence that supplementation improves memory or prevents cognitive decline. Some observational studies have suggested a correlation between higher niacin intake and reduced risk of Alzheimer's or cognitive impairment, but these findings are not consistent and may be confounded by overall diet quality. Small clinical trials exploring niacinamide for Alzheimer's disease have not demonstrated clear benefit.
