D-Aspartic Acid (DAA) is an amino acid that has been investigated for its potential effects on the male reproductive system, primarily regarding testosterone production and fertility. Scientific interest in DAA arose from early studies suggesting it may play a role in the synthesis and release of hormones such as luteinizing hormone (LH) and testosterone. Several small-scale clinical trials have explored DAA supplementation in men. Some studies, such as a 2009 study by Topo et al. (Reproductive Biology and Endocrinology), found that DAA supplementation (3g/day for 12 days) led to modest increases in testosterone levels in healthy men and those with low sperm count. However, subsequent research, including randomized controlled trials in healthy, resistance-trained men, have generally failed to replicate significant increases in testosterone, with some studies showing no effect or even a decrease after longer supplementation periods.

Currently, the evidence supporting DAA's effectiveness for boosting testosterone or improving fertility in healthy men is inconsistent and limited. Some effects may be seen in men with subfertility or low baseline testosterone, but these findings are not robust. There is no strong traditional use of DAA for male reproductive health; its use is based on relatively recent scientific inquiry. Thus, while DAA is sometimes used in supplements marketed for male reproductive support, its efficacy is not well established, and more high-quality research is needed.
