Vitamin D has some scientific basis for its use in supporting intestinal barrier function, which is often referred to in the context of "Leaky Gut Syndrome" (increased intestinal permeability). Several experimental and clinical studies have explored the role of vitamin D in gut health. Vitamin D receptors (VDRs) are expressed in intestinal epithelial cells, and activation of these receptors has been shown to help maintain the integrity of tight junctions—protein complexes that regulate the permeability of the intestinal barrier.
Animal studies suggest that vitamin D deficiency can disrupt gut barrier function, leading to increased permeability. Some in vitro and animal research indicates that vitamin D supplementation may restore or preserve the function of tight junction proteins such as occludin and claudins, reducing the risk of unwanted particles and bacteria crossing the gut wall. In humans, observational studies have found associations between low vitamin D status and increased gut permeability, as well as higher rates of gastrointestinal inflammatory diseases (e.g., inflammatory bowel disease).
However, there is limited direct clinical evidence specifically supporting the use of vitamin D supplementation to treat "Leaky Gut Syndrome" as a standalone condition in humans. Most studies focus on related disorders or use surrogate biomarkers. As a result, while mechanistic and preliminary human data are promising, robust randomized controlled trials in this specific context are lacking, warranting a moderate but not high evidence rating.
