Bacillus subtilis natto is a spore-forming probiotic bacterium, traditionally used in the fermentation of natto, a Japanese food. Its use to support or treat "Leaky Gut Syndrome" (also known as increased intestinal permeability) is based on emerging, but still preliminary, scientific evidence. Several animal studies suggest that B. subtilis supplementation can enhance gut barrier integrity, modulate immune responses, and promote the growth of beneficial gut microbes. For example, some research in rodent models has shown that B. subtilis can upregulate tight junction proteins and reduce inflammation in the intestinal lining, factors considered relevant to leaky gut. However, direct clinical evidence in humans with diagnosed increased intestinal permeability or “Leaky Gut Syndrome” is extremely limited. The majority of human research has focused on general gastrointestinal health, not specifically on leaky gut or its clinical consequences.

Systematic reviews of probiotics for gut barrier function often mention Bacillus subtilis as a promising candidate but emphasize the need for larger, controlled human studies. Therefore, while there is a plausible scientific rationale and some supporting preclinical data, the evidence for B. subtilis natto in treating or preventing leaky gut in humans remains weak to moderate (rated 2/5), and it should not be considered a proven therapy for this condition at this time.
