Bacillus subtilis is a spore-forming probiotic bacterium that has been investigated for its effects on gut health, including the integrity of the intestinal barrier. The rationale for its use in Leaky Gut Syndrome (also known as increased intestinal permeability) comes primarily from preclinical research and limited human studies. Animal models suggest that B. subtilis can upregulate the expression of tight junction proteins (e.g., occludin, claudin), reduce inflammation, and modulate the gut microbiota composition, all of which may contribute to improved gut barrier function. Some small-scale human trials and case studies have shown that spore-forming probiotics, including B. subtilis, may reduce gut permeability and symptoms associated with irritable bowel and other gastrointestinal disorders.

However, robust, large-scale clinical trials specifically assessing B. subtilis for "Leaky Gut Syndrome" in humans are lacking. The evidence supporting its use for this condition is therefore preliminary, and most claims are extrapolated from animal studies or from indirect evidence in related gut disorders. While the mechanistic rationale is promising, and the bacterium has a good safety profile, more rigorous research is necessary to definitively validate its efficacy for leaky gut in humans.
