Bacillus species, particularly Bacillus coagulans and Bacillus subtilis, are spore-forming probiotics that have been investigated for gut health, including their potential role in supporting "Leaky Gut Syndrome" (increased intestinal permeability). There is a growing body of preclinical evidence (animal and in vitro studies) suggesting that Bacillus probiotics can help modulate the gut microbiota, enhance the intestinal barrier function, and reduce inflammation. For example, some studies have shown that Bacillus subtilis can upregulate the expression of tight junction proteins in intestinal epithelial cells, which are critical for maintaining barrier integrity. Other research indicates that Bacillus strains may reduce pro-inflammatory cytokines and oxidative stress, both implicated in increased gut permeability.

However, direct clinical evidence in humans for the efficacy of Bacillus species in treating or preventing "Leaky Gut Syndrome" is limited. Most human studies with Bacillus probiotics have focused on general digestive health, irritable bowel syndrome, or antibiotic-associated diarrhea, not specifically on leaky gut. The evidence supporting their use for leaky gut is therefore considered preliminary and indirect. As a result, while there is some scientific rationale and emerging evidence, robust clinical validation is lacking at this time, resulting in a moderate-low evidence rating (2/5).
