L-proline is a non-essential amino acid that plays a crucial role in the synthesis of collagen, a major structural protein found in connective tissues, including cartilage, tendons, and ligaments that support the joints. The rationale for using L-proline to support joint health is primarily based on its biochemical role as a precursor for collagen formation. Several in vitro and animal studies have demonstrated that proline availability can influence collagen synthesis and deposition. However, the direct clinical evidence supporting supplemental L-proline for improving joint health or alleviating joint-related conditions in humans is limited and not robust.
Most of the scientific literature focuses on the importance of a combination of amino acids, including proline, glycine, and lysine, for optimal collagen production, rather than proline alone. Human studies on oral proline supplementation for joint health are scarce; most clinical research uses collagen hydrolysate or gelatin, which contains proline among other amino acids. Therefore, while there is a scientific rationale based on biochemistry and some preclinical evidence, high-quality human trials demonstrating direct benefits of L-proline supplementation for joint health are lacking, resulting in a moderate evidence rating.
