The largest and most rigorous trials, such as the GAIT trial funded by the National Institutes of Health, found that glucosamine (alone or with chondroitin) was not significantly better than placebo for pain relief in most participants, though a subset with moderate-to-severe pain did appear to benefit.

Systematic reviews and guidelines, including those from the American College of Rheumatology and the Osteoarthritis Research Society International, generally conclude that the evidence for glucosamine is mixed and does not robustly support its routine use for joint health.
