Cholic acid is a primary bile acid synthesized in the liver from cholesterol and plays a crucial role in the digestion and absorption of fats and fat-soluble vitamins. Its therapeutic application in liver and bile-related disorders, including certain forms of jaundice, is supported by scientific evidence—though this is limited to specific etiologies. Cholic acid is approved for the treatment of bile acid synthesis disorders due to single enzyme defects (such as 3β-hydroxy-Δ5-C27-steroid oxidoreductase deficiency and Δ4-3-oxosteroid 5β-reductase deficiency), which can manifest as cholestatic jaundice in both children and adults. By supplementing endogenous cholic acid, negative feedback on cholesterol 7α-hydroxylase is restored, reducing hepatotoxic metabolite accumulation and improving bile flow, which can alleviate jaundice in these rare conditions.
