Ursane-type triterpenoids, such as ursolic acid and corosolic acid, have been studied in the context of Inflammatory Bowel Disorders (IBD), including ulcerative colitis and Crohn's disease. Preclinical studies, primarily in animal models and in vitro systems, have demonstrated that these compounds possess significant anti-inflammatory and antioxidant properties. Mechanistically, ursane-type triterpenoids are known to inhibit the NF-κB signaling pathway, reduce pro-inflammatory cytokine production (such as TNF-α, IL-1β, and IL-6), and attenuate oxidative stress. Some studies have shown that administration of ursolic acid can ameliorate colitis symptoms in experimental mouse models by decreasing inflammatory cell infiltration and improving histopathological scores. However, evidence in humans is limited. Most data come from animal studies or cell culture experiments, with a lack of robust clinical trials directly evaluating the efficacy and safety of ursane-type triterpenoids in patients with IBD. Nevertheless, the consistency of beneficial effects observed in preclinical studies suggests potential therapeutic value, justifying further investigation. In summary, while there is scientific rationale and promising preclinical evidence supporting the use of ursane-type triterpenoids for IBD, the clinical evidence is currently insufficient. Thus, their use is justified by scientific understanding at the preclinical level, but rated as moderate (3/5) due to the lack of direct human data.
