Piperine, an active alkaloid found in black pepper (Piper nigrum), has been studied for its anti-inflammatory and antioxidant properties, which may be relevant to Inflammatory Bowel Disorders (IBD) such as Crohn's disease and ulcerative colitis. Preclinical studies, primarily in animal models, have demonstrated that piperine can reduce markers of inflammation in the gut, including decreased levels of pro-inflammatory cytokines (e.g., TNF-α, IL-1β) and mitigation of tissue damage in chemically-induced colitis. Some studies suggest that piperine may modulate the NF-κB signaling pathway, which is central to inflammatory responses. Additionally, piperine has been shown to enhance the bioavailability of other therapeutic compounds, which could indirectly benefit IBD treatment. However, there is a lack of robust clinical trials in humans investigating piperine as a standalone or adjunct therapy for IBD. Most evidence comes from in vitro and animal research, and there is little documentation on traditional use of piperine or black pepper specifically for IBD. Thus, while there is a scientific rationale and some supporting preclinical data, clinical validation is limited. The current evidence level is best rated as low to moderate (2/5), indicating that further research, especially human studies, is needed to confirm efficacy and safety.
