Casticin is a flavonoid compound found in various plants, most notably Vitex agnus-castus (Chaste tree). Its use for Inflammatory Bowel Disorders (IBD), which encompass Crohn’s disease and ulcerative colitis, is primarily supported by preclinical scientific research rather than traditional medicine. Several in vitro and animal studies have investigated casticin’s anti-inflammatory properties. For example, research has shown that casticin can inhibit the activation of NF-κB, a transcription factor involved in the expression of pro-inflammatory cytokines that play a key role in the pathogenesis of IBD. Studies in mouse models of colitis have demonstrated that casticin administration can reduce colon inflammation, decrease myeloperoxidase activity, and lower levels of inflammatory mediators such as TNF-α, IL-1β, and IL-6. However, these findings are limited to laboratory and animal models; to date, there are no published human clinical trials evaluating casticin for IBD. Therefore, while the current evidence suggests potential therapeutic effects via anti-inflammatory mechanisms, the evidence strength is rated 2/5: promising in preclinical settings but lacking direct clinical validation. Casticin is not established in traditional herbal medicine systems specifically for IBD, and its use for this purpose is grounded in emerging scientific rather than historical evidence.
