Siphoviridae is a family of bacteriophages (viruses that infect bacteria), and their use in treating bacterial infections is supported by a growing body of scientific evidence. Bacteriophage therapy, including the use of Siphoviridae phages, has been explored for over a century, but recent advances in molecular biology and genomics have revitalized interest in their application as alternatives or adjuncts to antibiotics, especially in the context of antibiotic-resistant bacterial infections.

Numerous in vitro and in vivo studies have demonstrated the ability of Siphoviridae phages to specifically target and lyse pathogenic bacteria such as Staphylococcus aureus and Escherichia coli. Several case reports and small clinical studies have reported successful treatment of multidrug-resistant infections using phage preparations containing Siphoviridae members. The specificity of these phages reduces the risk of disrupting normal microbiota compared to broad-spectrum antibiotics.

While large-scale randomized controlled trials are still limited, the scientific foundation for the use of Siphoviridae in phage therapy is strong, supported by mechanistic studies, compassionate-use cases, and early-phase clinical work. This places the evidence at a moderate-to-high level (4/5), pending further clinical confirmation. Regulatory, manufacturing, and safety considerations remain, but the scientific rationale for using Siphoviridae against bacterial infections is well-established and growing stronger with ongoing research.
