Glutathione peroxidase (GPx) is a critical antioxidant enzyme that plays a major role in protecting the liver (hepatic system) from oxidative stress. This enzyme catalyzes the reduction of hydrogen peroxide and organic hydroperoxides, using glutathione as a substrate, thereby neutralizing potentially harmful reactive oxygen species (ROS) that can cause cellular and tissue damage. Scientific studies have demonstrated that the liver is especially susceptible to oxidative damage due to its role in detoxification and metabolism of various substances, which often generate ROS as byproducts. Decreased GPx activity has been observed in a variety of liver diseases, including non-alcoholic fatty liver disease, alcoholic liver disease, and hepatitis. Conversely, upregulation or supplementation of GPx has shown protective effects in animal models and some human studies, reducing liver damage markers and improving liver function. Clinical research supports the notion that enhancing GPx activity, either directly or by increasing levels of its cofactor glutathione, can mitigate oxidative stress and support hepatic health. While most interventions focus on glutathione precursors or selenium (a cofactor for GPx), the centrality of glutathione peroxidase in hepatic antioxidant defense is well established in the scientific literature.
