Nicotinamide Mononucleotide (NMN) is a precursor to nicotinamide adenine dinucleotide (NAD+), a vital coenzyme involved in cellular energy metabolism and redox reactions. Scientific interest in NMN has grown due to its potential role in mitigating free radical damage, a process implicated in aging and various diseases. NAD+ plays a central role in cellular antioxidant defenses by serving as a substrate for sirtuins and PARPs, enzymes that regulate oxidative stress responses and DNA repair. Preclinical studies in mice and cell cultures have shown that NMN supplementation can increase NAD+ levels, enhance the activity of antioxidant enzymes, and reduce markers of oxidative stress. For example, NMN has been reported to reduce lipid peroxidation and DNA damage in animal models exposed to oxidative stressors. These findings suggest a mechanistic basis for NMN in supporting the body’s defense against free radical damage. However, direct clinical evidence in humans is limited. Most human trials to date have focused on NMN’s safety and its effect on NAD+ levels rather than its efficacy in reducing oxidative stress or free radical damage. While the biochemical rationale is strong and animal research is promising, large-scale, well-controlled human studies are needed to confirm NMN's effectiveness for this specific indication. Therefore, the current scientific evidence rates as moderate (3/5) for NMN’s use in supporting or treating free radical damage.
