Rhaponticin is a stilbene compound found primarily in the roots of Rheum rhaponticum (rhapontic rhubarb). Recent preclinical studies have investigated its effects on fatty liver disease (hepatic steatosis), particularly in animal models. Evidence suggests that rhaponticin exhibits hepatoprotective properties, including the reduction of hepatic lipid accumulation and improvement of liver enzyme levels. Mechanistically, these effects have been attributed to its ability to modulate lipid metabolism, reduce oxidative stress, and suppress inflammatory pathways in the liver.
For example, a 2016 study in mice fed a high-fat diet demonstrated that rhaponticin supplementation reduced liver triglyceride content and improved histological markers of steatosis. The compound was also shown to regulate genes involved in lipid synthesis and breakdown, such as SREBP-1c and PPARα. Additionally, rhaponticin displayed antioxidant activity by increasing superoxide dismutase (SOD) levels and decreasing malondialdehyde (MDA), both markers associated with oxidative liver damage.
However, most of the available evidence comes from animal studies and in vitro experiments; there are currently no published clinical trials in humans directly evaluating rhaponticin for nonalcoholic fatty liver disease (NAFLD) or related conditions. Thus, while preliminary findings are promising, the scientific validation remains limited and further research, especially in humans, is required before definitive conclusions can be drawn regarding its efficacy and safety.
