Nicotinamide Mononucleotide (NMN) is a precursor to NAD+ (nicotinamide adenine dinucleotide), a molecule involved in metabolic and cellular processes, including energy metabolism and mitochondrial function. Fatty Liver Disease, including non-alcoholic fatty liver disease (NAFLD), is characterized by the accumulation of fat in liver cells and is linked to metabolic dysfunction. Some preclinical studies have suggested that boosting NAD+ levels through NMN supplementation may help improve hepatic lipid metabolism, reduce inflammation, and enhance mitochondrial function, potentially benefiting liver health.
Animal studies have demonstrated that NMN administration can reduce hepatic fat accumulation, improve insulin sensitivity, and decrease markers of liver inflammation in models of diet-induced fatty liver disease. For example, research in mice has shown that NMN supplementation can ameliorate hepatic steatosis and improve overall liver function. However, human clinical trials are limited. The existing evidence in humans is preliminary and largely indirect, with only a few small studies examining NMN’s metabolic effects, and none providing definitive proof of benefit specifically for fatty liver disease.
In summary, while there is promising preclinical evidence and a plausible mechanistic rationale for NMN use in supporting fatty liver disease, high-quality clinical trials in humans are lacking. Thus, the current evidence is rated as low (2/5), and NMN should not be considered a validated treatment for fatty liver disease at this time.
